Western blot was performed using Anti-TAP Tag Monoclonal Antibody (22F12-1E3) (Product # MA1-108) by loading whole cell extracts of untransfected and transiently transfected HEK-293E lysates: untransfected, 50 µg (Lane 1), empty vector control, 50 µg (Lane 2), H3-TAP, 50 µg (Lane 3), H3-TAP, 25 µg (Lane 4), H3-TAP, 12.5 µg (Lane 5), H3-TAP, 6.25 µg (Lane 6), p65-TAP, 50 µg (Lane 7) were electrophoresed using NuPAGE™ 4-12% Bis-Tris Protein Gel (Product # NP0322BOX). Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). A ~36 kDa band corresponding to H3-TAP and ~81 kDa band corresponding to p65-TAP were observed in HEK293E transfected lysates on probing with the primary antibody (1:1000) and detected by chemiluminescence with Goat anti-Mouse IgG (H+L) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A28177,1:4000) using the iBright FL 1000 (Product # A32752). Chemiluminescent detection was performed using Novex® ECL Chemiluminescent Substrate Reagent Kit (Product # WP20005).
